The heater unit under my dash shows two knobs (49 deluxe), one air flow and one defroster. The air flow is working. The defroster not. Can anyone tell where this defroster knob is for and how it works? Thanks

I would think that the other control goes to a water valve. You may be able to trace the cable to the valve.

On the heater controls shown below, the knob on the left controls the speed of the heater motor and the knob on the right controls the speed of the defroster motor.

OK, but this defroster motor? Where is it situated, and what does it do? Where do I have to use it for? If I turn this one on, do I have to hear something or not? Thanks.

The heater and defroster blower motors on a 1949 DeSoto are located within the heater/ductwork unit under the hood on the passenger side as shown below. If the defroster is working properly, it should be blowing air out onto the inside of your windshield from the base of the windshield when on. This would be used to keep condensation from forming on the inside of the windshield and to help melt frost or snow on the outside. At high fan speed, you would probably hear the fan running.
